The 22-year-old striker found the net twice within the opening 17 minutes at the Metropolitano Stadium, ensuring Atletico’s place in the last four for the second consecutive season.
Samuel Lino extended the hosts’ lead with a well-taken strike late in the first half, while Ángel Correa and Alexander Sorloth added two more goals in the second half to seal the emphatic win.
Atletico, who last lifted the Copa del Rey in 2013, fell short in last year’s edition after being knocked out by eventual champions Athletic Bilbao.
Elsewhere in the competition, Real Madrid will face Leganes on Wednesday, while Thursday’s action features Real Sociedad hosting Osasuna and Barcelona traveling to take on Valencia.
Giuliano opened the scoring for Atletico with a powerful header in the eighth minute and soon doubled the advantage after finishing off a breakaway in the 17th.
Lino made it 3-0 in the 42nd with a clever move to beat a defender before curling a low shot into the far corner.
Correa added a fourth with a long-range effort in the 78th minute, and Sorloth capped off the victory with a clinical finish in the 86th.
With a crucial La Liga clash against league leaders Real Madrid looming on Saturday, coach Diego Simeone rotated his squad, giving several fringe players a chance to impress.
Atletico currently sits just one point behind Madrid in the league standings.
Getafe, meanwhile, managed only a single shot on target in the first half and continued their poor form, with just one win in their last four matches across all competitions.
The club has not reached the Copa semifinals since the 2009-10 season.
